The first reading is determining your sand, and that should mostly settle out by the first reading. The second reading for clay is needed just to determine how much silt is in the sample. If all you are after is an estimation of soil texture it would be less time consuming to hand texture your samples. I'm not sure what effect a 250ml cylinder would have if any on the settling of your sample, and you'd have to adjust everything else. The 250ml cylinder may not allow enough space for the hydrometer.

I have question, can I use the 250 ml measuring cylinder rather than 1000ml ? And when I have little silt+ clay mixture how should proceed it? Thanks
