Wake Forest University is in Winston-Salem, NC. It is named for the town of Wake Forest, NC, which is where it was originally founded. That town is just north of Raleigh, NC.
Missed a few important details. Both Wake and Duke were originally tobacco manufacturers, but like warring tribes, Duke forced Wake out, and both started their own universities.
Fun fact: Wake Forest (judging by total student population including undergrads and post-grads) is the smallest power-5 school in the country. It’s a private school (very expensive just like it’s in-state counterpart Duke) it’s in the ACC conference and has one of the most Incredibly Beautiful campuses I’ve ever seen. I’m biased since I got my undergraduate degree there…but left and got my Graduate Degree from the University of North Carolina @ Wilmington (which is also a great school). But damn I missed those power-5 football/basketball games no doubt during the two years I spent at UNCW getting my Grad degree.

The founder of RJ Reynolds Tobacco Company (the make Camel and almost everything else) wanted his daughter to go there but he didn't want her to be so far from home. So he FREAKING PAID for the university to build a whole new campus and move to Winston, and they've been here ever since.
Mr Reynolds’s also just wanted a big university in Winston so then the city would become more educated. He brought the college to Winston not just for a personal reason but also for the benefit of the city.
Another Winston-Salem native and lifelong Wake fan here. The WFU campus being moved from Wake Forest to Winston-Salem actually nothing to do with R.J. Reynolds Sr. himself, since he died over thirty-five years before the move even occurred. The sale of the original 330-acre estate for the Winston-Salem campus was actually put forth by one of RJR’s daughters, Mary Katharine, and her husband, Charles Babcock. The move was largely done as an act of general philanthropy for the city of Winston-Salem, but there is some important time-period specific cultural context to the move as well. The move was also made to give the R.J. Reynolds Tobacco Company a resource for hiring top-tier students/future business talents to eventually run and take over the day-to-day operations of the company, in very much the same way that the Duke Endowment/identity change of Trinity College to Duke University had essentially done the something for the American Tobacco Company (RJR Tobacco’s primary business rival) based out of Durham.
