So backlog is the Issue Status category ? I work with Jira data import and try to find clear programming creteria how to find issues in the backlog, Can issue in backlog has assignee ? or any Sprint assigned ?

Thanks for this video! Should designs be done before or after a ticket is refined? Also, should design tickets be sub-tasks or just linked to a story?

My opinion as there are no silver bullets here. If your designs are requirements, then they need to be done before refinement. I would prefer a story, because you can plan those into a sprint, but both techniques will work.

Is there any options to get email threads in jira tickets? As of now we need to copy paste further informations from email once after a ticket is automatically created. So I need to know Is there any way to automatically get the subsequent details in the jira ticket.

As long as the issue key is mentioned and the email for your jira is in the CC, those emails should continue showing up in your comments section.
