Without too many spoilers, his line claiming he's "one of the strongest in the tower" is not an exaggeration or bluff, it's genuine truth. Obviously taking into account the amount of people in the tower and the skills of some of the actual rankers we've seen in the anime it tells you everything you need to know that he's disposing of rankers with ease.

We've barely seen a handful of rankers so far and how strong they are compared to regulars, but to now include the #4 in the whole Tower without a proper introduction or understanding of his character can really confuse and take away the hype. The casual remark that Mazino is casually disposing of rankers can make it seem like rankers are weaker than previously thought rather than emphasising how strong Mazino actually is (for anime only watchers). For those who've read TOG it's hype but I think it could have been done better for anime only watchers.
Your memory is good Brian, Mazino was mentioned in the first season. As he said in the ep he's one of the most powerful people in the tower. I haven't been a fan of how they've animated the series so far, so hopefully this battle will live up to the hype 😁
