      haha that might be funny. Cadaverine wasn't that bad though because the vapor pressure isnt super high. The trimethylamine i made a week later was so much worse

      NileRed I think we need to see you react to some of your chemicals, even as a joke. if its safe to smell some of your concoctions, it would add a welcome human element to the presentation, at least for the casual viewers who aren't chem majors (like most of us!)

    I made a mixture of both when I forgot a crockpot full of pulled pork in my house before I went on a camping trip for a week. It was meant for the camping trip.. The smell was so bad that I instantly threw up when I encountered it, I decided to just throw out the entire crock pot.

    One thing I noticed is that your N-bromosuccinimide was very orange. NBS readily releases Bromine upon exposure to light so it must be kept in the dark. For reactions that uses the electrophilic properties of NBS, it is usually recommended to perform them in glassware wrapped in aluminium foil. To obtain pure NBS, you can recrystallise it very easily from 90°C water using 100ml of water for 10g of NBS.

    Please consider posting more of your longer projects. The longer the video, the better, I really find them relaxing to fall asleep to. I’m an extreme insomniac, and have a really hard time going to sleep, and your videos really really help. I’ve watched all your long videos, and most of your short ones. I’ve watched almost every video on this channel and your other one. the videos that are 20mins+ are the ones I’ve found to be the best. Anything less than that ends up with me to play 3-4 videos before I fall asleep, and ive pretty much watched every videos several times now

    I honestly find it very Interesting to see things like these are possible, since it is so different from what we made at school and (might be because of my very limited chemistry skills) I never thought of those possibilities.
    I also have to add that I learned more chemistry by watching Nile Reds vids, then by attending 3 years of Chemistry in school...

    About the reaction, water will slowly decompose your sodium borohydride, so make sure to use a reasonable excess. Secondly, try to vacuum distill instead of extract. After that, you can evaporate the water away. What's left will be your crude product.

    At least in the case of L-phenylalanine, an amino acid can be converted to its corresponding nitrile (with one less carbon atom, just as you do in your process) using the slow addition of household bleach at low temperature. I've used this procedure for preparing benzyl cyanide from phenylalanine.

      Hi Nile, better descarboxilate amino acids with TCCA (trichloro cyanuric acid - pool shock) to get one less carbon nitrile with better yields. See:

      This way you just need to filter the insoluble cyanuric acid to obtain just nitrile and water.
      You'd better use methanol as the solvent in the nitrile reduction with nickel boride and mantain the temperature always below 10ºC.

      The use of Dimethylformamide (DMF) as you did can result in their reduction with the formation of trimethylamine and dimethylamine as a byproduct and contaminant hard to separate from cadaverine.
      I think the experiment like you did can't be done as an one pot reaction, otherwise this explain the poor yield you've got.
      Since you asked for sugestions, mine goes this path: reduce lysine with TCCA --> filter the cyanuric acid precipitaded --> extract the nitrile from water with dichloromethane (DCM) --> dry the solution with calcium chloride or anhydrous magnesium sulfate --> add a methanolic soluction of nickel chloride hexahydrate --> freeze the solution to 0ºC --> add sodium borohydride in portions and slight excess -->filter the nickel boride --> add 10% sulfuric or chloridric acid to make free the amine salt (water soluble) and neutralize the excess borohydride --> wash with DCM again to extract the unreacted nitrile and discard --> basify with 6M NaOH to liberate the amine --> extract with ether and evaporate.

    I've had similar problems with partition coefficients going the wrong way with a couple of reactions in my scheme. I would recommend using a more polar solvent like ethyl acetate instead of ether and doing something like ten extractions. You could then reextract it from the organic layer with an aqueous acid solution, wash with ether to remove organic impurities, make it alkaline and salt out the product with brine.
    That was a nice demo of a pretty uncommon reducing agent. Thanks!

    Good job! having read the paper I was wondering if this reaction could be applicable to Trp and Tyr; I also think the reaction is borohydride wasteful but there's some old one I saved from throwing away at school so I can just add until it reduces all the substrate... I'm not sure if Ni sulfate can be a viable substitute for the chloride though

    Regarding the nickel boride removal step, do you have access to celite? Its really good at getting rid of metals. I use it to remove tin (II) chloride and silver perchlorate.

    This is a cool video and all, but something in me feels like this was a waste of chemicals. (Kids in Africa could have eaten that sodium borohydride!) By that, I mean that three are cheaper reagents you could have used to perform the same reaction. TCCA in sodium hydroxide is a known reagent to convert amino acids and amines to nitriles ( this would instead give you the dinitrile), and CTH with a tiny amount of palladium catalyst would reduce this to the diamine easily. This follows the same synthetic pathway, but is much more reagent efficient.
    Of course, this also neglects direct decarboxylation: although straight heating is a poor method, decarboxylation of amino acids in acetophenone is known to proceed in high yield and at temperatures under 150 C, due to the formation of an imine intermediate which is easier to decarboxylate.

      I think it can be re-used as a catalyst so we dont have to make it in situ the next time. I am not 100% sure though.
