If you have US Passport you are 100% free to leave the airport and roam around London, no Visa needed. The process is simply the same as going into a country go through customs and show your Passport and they'll let you go.

I would recommend checking on the internet for that question, because I don't want to give you wrong infromation, but i can tell you 6 hours in plenty of time to go into the city and take a picture of Big Ben and the because the train will take you very close to there and you'll have time to make it back on the train to the airport. Every country has their own policy for example China allowed me to get out with no Visa to visit Shanghai but Jordan did not allow me to do it without visa. I would suggest to check with the embassy policy
