    The CelCradle™ operates through the Tide Motion principle wherein cells, attached to BioNOC™ II carriers, are alternately exposed to aeration and nutrition via the decompression and compression of the bellows holding the culture medium. The gentle vertical oscillation of the culture medium creates a dynamic interface between air and culture medium on the surface of the cells, providing the cells with an environment that is of low shear stress, high aeration and nutrition levels, zero foaming, and no O₂ limitation. This efficient nutrient and oxygen transfer allows the CelCradle™ system to produce high cell density yield.
    The following video will show how to seed adherent cells in the CelCradle™ system.
    In the Biosafety cabinet, prepare seed in 120ml of fresh and pre-warmed culture medium.
    Ensure pH is between 7.2-7.4.
    Dispense seed mixture on the carriers.
    Place the blue vented cap in a sterile petri dish and cover the dish to ensure the sterility of the cap for future use.
    Cap bottle with non-vented white cap.
    Invert bottle.
    Ensure that all the 120 mL seed mixture drains down from the bellows to the cap. Make sure that the carriers are submerged in the seed mixture.
    Swirl well to ensure cells are spread evenly.
    Move the bottle into the CO₂ incubator.
    Allow cells to attach for up to 4 h.
    Swirl the bottle gently every 15 min to allow cells to be redistributed.
    At the end of the incubation period, remove the bottle and sample 10 ml seed mixture to check the number of cells unattached.
    Alternatively, check for the number of cells attached by harvesting cells from 3 carriers.
    Top up media in a bottle to 500 ml.
    Cap the bottle with the vented blue cap. The non-vented white cap should be left in the sterile petri dish and covered to ensure its sterility for further use.
    Move the bottle into the CelCradle™ Stage. Set up Tide Motion parameters and press “Start”.
    Parameters can be adjusted according to cell line characteristics and media conditions.
