It's all because of Ayanokoji mega big brain moves that ryuuen can't say for sure that Ayanokoji is the mastermind although he seems to fit the bill as the mastermind in class D. Every actions and decisions that Ayanokoji have done so far has a reason. For example, the race with Horikita Manabu. He didn't just flex his strength to the whole school just for show, part of it is to gain interest from Horikita's brother, but most importantly is to confuse ryuuen. Ryuuen thought that the mastermind wouldn't show them self easily so it was a strategy to divert ryuuen's attention to someone else. Also from this episode, the part when ryuuen and his underlings make an appearance in class D and went after chad Koenji. Hirata was gonna go along with Horikita to see the whole shit that was about to happen. But then Ayanokoji told him to stay cuz if he followed them, others like ike and yamauchi aka the trouble makers would follow to. That was part of the reason why Hirata shouldn't go but mainly was to confuse ryuuen into thinking that it may be Hirata as the mastermind cuz Ayanokoji knew it was a strategy by ryuuen to figure out the mastermind. He thought that the mastermind would not show himself when the whole shit was happening. And seeing Ayanokoji there makes it hard to deduce him as the mastermind cuz what Ayanokoji has done so far contradicts the action of what the mastermind would have done, which is to keep a low profile. Thus, the reason why it seems like ryuuen didn't supect or investigate Ayanokoji, tho he did suspected Ayanokoji just that he couldn't find any concrete evidence that it's him(cuz Ayanokoji is too smart for him)and simply making a wild guest that Ayanokoji is the mastermind simply because he seem to fit the bill as THE mastermind is not a valid option. In ryuuen's pov, there are a few potential mastermind candidate that he need to consider. From our pov as the audience, we knew all along it was Ayanokoji so it was easy for us to say that it was him, but not for ryuuen. To him, it might be a ploy set by the mastermind to confuse ryuuen from figuring out the identity of the real mastermind. My theory is that she's trying to: A) Reinvent herself and wants to distance herself from her past as much as possible and more people knowing about her past makes it harder for her to distance herself from it. and B) She's trying to avoid looking like an easy target to other potential bullies. If another bully heard about her past they could think something like "Another person pushed her around for years and she never did anything about it, she probably won't stand up to me either." No idea if i'm right, but it's the best reason I've come up with so far.

Revealing this past would give a weapon to those who hate her, and make her come back to the bullying victim she was. Bullying is still active, and Karuizawa and her friends actually bullied Sakura for quite a bit before Ayanokoji told her to stop it

So the LN or the manga explains why it is so hard for him to make that deduction... The anime cuts all that stuff out.
