Can we directly use this client Id and key in S/4 Hana Rfc Destination or we have to use only BTP USER ONLY

No I haven't tried using the OAuth Authentication, it is either with Basic Authentication or Logon ticket. This is to connect to your SAP BTP Integration Suite. I would advice you to go to the help on this transaction SM59 to understand the difference for logon ticket implementation. Since this is for your integration flow, a technical user should be enough to get the data from S/4HANA to you BTP Integration suite.

A user that has the right on your SAP BTP integration suite system.
What will be the host ? is it cpi tenant url or url we willget after configuing the iflow
