I think we're going to see Niacin as an increasingly viable option to its metabolites. I would put a small personal anecdote forward for those of us who get aura migraines; I have tried 500mg Niacin with TMG many times and found a growing incidence of aural migraine triggered after taking that size of dose. At this stage, if I take 500mg nowadays it's a guaranteed migraine. However, I dropped to 100mg and that has been fine for me. Just keep a track on dose and side effects after flushing.
Interesting. Just checked my spreadsheet. The response rate to NR (in blood; not sure of duration) was 138% (1.38x) @ 1,000 mg in an NR study (Niagen?). In this video's study, the response rate was 1.3x (very close) at 4 months, and 2.3x in 10 months (NR study didn't run this long). Understanding there's a difference between muscle and blood NAD levels, it would appear that Niacin is a viable alternative (caveated with its side effects, of course). Could they be used together to increase NAD from two different directions? I used the response rate to NR (mg vs %chg) from the "138%" NR study, and the fact that NAD levels fall "50% every 20 years", to create a calculator (2 regressions). Plug in your age and your desired age's NAD level, and out pops mg of NR required. Example (my Dad): Target Age: 68 Current Age: 81 mg NR needed: 397

Once that's done, I'm switching back to Niacin and NMN, and will probably end up with just Niacin because it's cheap and effective, and has positive side effects for me (lower LDL, higher HDL). I don't take a crazy dose, just enough to stay topped off. (250 mg time released)

I might add that I am now using a 'wax-matrix' niacin (nicotinic acid) that is slow release by the nature of the wax slowly dissolving. It is called Enduracin. I have taken up to 750 mg at a time, with no flushing.
I limit my dose of niacin. Please do a search of the key-words niacin macula. Recent papers indicate that there might be impact on the macula. And this study had 5 patients.

Niacine will give you a huge rush. When i take niacin in high doses all my skin gets red and itchy (its a common side effect), so not sure you can take a lot of it.

The trial lasted around 4-5 years but when they did a follow up a decade later they found a significant reduction in all cause mortality in the niacin group, in of the trial having been ended and the group had stopped taking niacin
I take 1500mg nicotinic acid a day, split into three 500 mg doses. Other than flushing, I've never experienced any other negative side effects... so no GI upset or flatulence, and a recent blood test put my haemoglobin and red blood cell count in the upper portion of the normal range.... I am only one data point, so take with a grain of salt, but the point is that these side effects are obviously not universal. O think it may be important to note that I also do a lot to support my liver health along side the niacin intake. I also ensure i get at least 600mg of choline, paired with inositol, as well as 1000 mg of TMG (Despite the fact TMG is a metabolite of choline, I choose to use both instead of just choline as tmg raises tmao levels less than choline). Plus i make sure to get ample b12 and folate plus a balanced amount of the other "B" vitamins to ensure sufficient methyl group intake and sufficient vitamin c, k1 and k2, d3, Magnesium, etc... but most importantly, along with the glycine from TMG and choline, i also take NAC and whey/collogen protein to supply sufficient L-cysteine to support glutathione levels.... another very important consideration when discussing liver health.
Be careful with niacin. My doctor prescribed some for lowering cholesterol. I took one pill. The side effect listed is flushing. That doesn't sound bad does it? An hour or so later I felt like my arms were on fire. After another hour it let up for a while and then started over again. I was ready to go to the emergency room when it finally quit.
High dosage niacin improves blood circulation hence the tingling and flushing . But it increases Uric Acid . The trial should have had all subjects aged over 65 . 17 is pretty pointless as musculature is still growing due to IGF .
The tingling and flushing is nothing to do with blood flow. Niacin liberates histamine and causes mast cell activation.the tingling is from prostaglandins being released into the bloodstream. It also only has a half life of about 45 mins so superdosing it in one sitting doesn't make sense. People less sensitive to histamine and mast cell activation will experience less tingling, but there is an upper threshold for everybody where they will all get the same outcome
A comment on NIACIN as a NAD+ precursor: I have worked up to 1 gram doses of NIACIN with a slight flushing for 20-30 seconds. However I have stopped a few days because of severe gout (my family has a history of gout, and I am 81 YO.) Research finds that NIACIN can result in gout. Looking at the Preiss-Handler pathway for NIACON conversion, I see TRYPTOPHAN involved. Maybe I should include TRYPTOPHAN?

You might want to study k2 for that as well since it does mobilize calcium in soft tissues Animal source k2 is the most bioavailable form but if you can stomach NATO (fermented soybeans) knock yourself out.

I'm a complete layman on the subject, but I know someone who developed mitochondrial myopathy since birth. Would niacin improve muscle performance for them or only people who develop it as adults?

I take niacin and I take the flushing type of niacin I take 3000 mg A-day for anxiety and depression and it works very well

Well these chemicals do set into motion different pathways. And for me, the reason I use both is that you can take NMN on an empty stomach.
