Apart from the time needed to constantly lift and drop it as needed, the problem is that it needs to be adjusted with the thickness of the section being cut to get the best result, which takes more time. Cryostat microtomy here is like the 'emergency ward' of the histo lab, so time is usually an important issue.
Sorry it's been a while Netra! We use Tissue-Tek OCT (optimal cutting temperature) compound. It's basically a very week solution of PVA with antifreeze in it (go figure hey!).

Different freezing agents are used including liquid gases like N2 at a temperature of -190 degree C ,liquid CO2 at a temp. of -70 degree C etc.
