I'm a huge fan, but have never given it a listen or the film a chance. My fave is Giu la Testa, Duck, You Sucker, and Fistful of Dynamite-same film with different release titles.
Just got the newly remastered La La Land release of "Sister Sara" in honor of Il maestro's passing. This is a really underrated work of his among his Western classics. Also gotta give Quentin Tarantino credit for bringing my attention to this score and film. If he hadn't used tracks 4 and 6 in "Django Unchained", I most likely wouldn't have ever seen this film or discovered this wonderful score. R.I.P. to Ennio.

Ennio Morricone's first score for an American film. This often gets lumped in with his spaghetti western scores although its not spaghetti or technically a western for that matter (Two Mules falls under the Zapatta clasification like Duck You Sucker).
@@humdunkin328 also didn't she swap to a smaller mule half way through the film when hers had something in its hoof ? and that one she didn't ride side saddle ?

If you are talking about "the braying mule" in opening theme, it is an early electronic keyboard called Clavioline.
