Materials Masterclass: Strong and fragile glass formers explained!
Lots of pharmaceuticals can form glasses if cooled rapidly, but some are described as strong glass formers and others as fragile glass formers. But what do these terms mean? Where do they come from? And should I worry whether my material is a strong or fragile glass former? All of these questions and more* will be answered in this video!
*Actually there's no more. That's yer lot :)

How can the same material have different glass transition temperatures?!
มุมมอง 208หลายเดือนก่อน
How can one compound appear to have different Tg values? That's crazy! Surely there should be only one, true value? Well, you'd think so but in reality the way the glass is made AND the way Tg is measured will all affect the Tg value determined. But why? What is it about formation that changes Tg? And why do different analytical techniques give different Tg values? All will become clear in this...
Materials masterclass: Plasticisers in amorphous solids (glasses)
มุมมอง 2462 หลายเดือนก่อน
Ever wondered why amorphous pharmaceuticals are so unstable when there's water around? It's because water is an excellent plasticiser, and speeds up the rate at which amorphous materials relax, ultimately leading to crystallisation. Plasticisers are also added to all sorts of plastics and polymers, to change their mechanical properties and feel to the touch. But what are they? And how do they w...
DSC Masterclass: How to interpret Glass Transitions
มุมมอง 3112 หลายเดือนก่อน
I can't tell you how good our students seem to be at *seeing* glass transitions in their materials when run in a DSC. But are they actually glass transitions or just desperation on the part of the students? Mmm... Here we look at how glass transitions appear in a DSC, why they should be seen as a step change in the baseline and why they are sometimes seen as an exothermic peak. By exploring why...
Understanding the science behind relaxation of amorphous solids
มุมมอง 2432 หลายเดือนก่อน
Amorphous materials are tricky! For one thing, they are always changing. If you leave candy floss (or cotton candy) in the open air it will have shrunk to almost nothing in just a few minutes. Or if you're a racoon and you put your cotton candy in water you'll be a sad and confused racoon (th-cam.com/video/rfbb4yRBH64/w-d-xo.html). Not good! But why? What is it that amorphous glasses do that me...
Revealed: Why Amorphous Solids DON'T Melt!
มุมมอง 4552 หลายเดือนก่อน
Amorphous solids are just that - solid. Well, below their glass transition temperature anyway. When heated they turn to liquids, so just like crystalline solids they melt, right? No! In fact an amorphous material CANNOT melt. So what do they do? Well, that's what we'll look at in this video. The glass transition temperature (or Tg) is one of those mysterious concepts that seems very hard to understand - not any more! The concept is highlighted with diagrams, video and an indecent amount of indomethacin. There are some related videos you might also wa...

    Melting point can not determine the Gibbs free energy directly. So sometimes, the higher melting point polymorph can be meta-stable form at certain temperature.

    As always, great video Dr. Gaisford! One question related to the video: when heating an aged glass sample just above the Tg, would the cooling rate matter in a similar way as if we were cooling directly from the melt? Also, could you make a video on modulated DSC and how it helps separate "reversing" and "nonreversing" signals? Even after watching lots of videos online, which just throw some equations at me, I feel like I don't really understand it. I love the way you teach, so thank you for taking the time out to make these videos!

    • @PharmaDrama
      @PharmaDrama 2 หลายเดือนก่อน

      Now you raise some very good points! Firstly, cooling rate would matter - the faster the cooling rate, the lower the Tg will appear. There's a video on that coming soon (the next video will be about plasticisers, and then cooling rate). I will indeed do a video on modulated DSC, although to be fair the maths behind that is tricky! Glad you're finding the videos helpful though :)

    Is relaxation a pre-requisite for crystallization? I think it should be because of the increasing density, but could you please confirm/explain? Great videos, Dr. Gaisford!

    • @PharmaDrama
      @PharmaDrama 2 หลายเดือนก่อน

      Thank you for your question :) I think the answer is yes, only because relaxation means the movement of molecules, and crystallisation requires the movement of molecules - hence they must go together.

    • @PharmaDrama
      @PharmaDrama 2 หลายเดือนก่อน

      Thank you for your kind words. Nicotine and cocaine are both called drugs because they are the chemical species that elicit a response in the body. If formulated they would be medicines. I think the confusion is that some drugs are subject to legal restrictions; for instance, controlled drugs, such as morphine, or illegal drugs such as cocaine. But to the general public the word 'drug' is often used only for illegal substances.

    if the solubility of the stable form is below industry standard, what to choose for formulation ? if you choose a metastable form, it might transition into a stable form with time. What is the best practice ?

    • @PharmaDrama
      @PharmaDrama 3 หลายเดือนก่อน

      Yes, that is a tricky question. I guess the answer depends on whether you get enough bioavailability for your active. If not then a metastable form is an option but I think most companies would go with the stable form and use an alternative solubilisation strategy (eg milling, salt formation or mixing in an amorphous polymer).
