Such a great scene. Correct me if im wrong, but I feel that L is pitying Light here too. L obviously is showing that he cares for Light, as a friend (I mean he says it before), but also at the same time he hates him cos he is Kira and him being Kira means either one falls, but since he cannot prove it, he is disappointed with not only himself, but especially with Light, cos a bright guy with the same intelligence as him (probably more) he decided to go the other way and he is sad about it. And with that question he just validated that Light has no morals, and he knows he will die trying to prove it quite literally in fact.

I mean, if I was L in this position, and I knew 99.9% certain that Light was Kira, wouldn’t it be better to have just killed Light on the rooftop? Yeah he’d be arrested (stupid I know) and viewed as a murderer, but Amane would just end herself, so one less notebook, and Teru is so incompetent they’d have no problem catching him, pretty sure it would all work out after right?

I've always wondered why L never did anything despite knowing light was Kira. Maybe L saw light as a friend and didn't care? Or was done with it all due to being alone and didn't care that light was Kira?

I have a tendency to come back to this scene every once in a while. It's just fascinating to me how with just audio effects, they have conveyed the entire emotional tension between Light and L. It's a layered question which in a way represents L's victory over Light. He's always known Light is Kira with increasing levels of uncertainty. The question may seem a bit of an exaggeration at first, I mean everyone had to have told the truth at some point on their life. But it never was about the answer. Beyond that surface, L is mocking Light with a statement, rather than a question. For as long as he has known Light, there were always lies between them. Everything was a lie; their initial contact, the investigation, the schemes and ultimately Light managed to convince the whole world (more specifically, everything surrounding L) with his lies. Everyone but L, believed it. But L saw through it all and in this scene he is mockingly pondering if Light has lied the whole time even before they met, thus extending his whole life. Light has to pretend he can only see the surface level and tries to answer the "right way" by diminishing the implication. But he knows that "good" answer is just another pretense as if he doesn't understand the deeper layers, thus proving L's point. He knows there's nothing he can do but to take the humiliation of taking the question seriously. He's forced into a checkmate so he just makes a vague generic answer. And L has completely lost interest in that. He said what he wanted to say and his opponent couldn't even face him properly at that last moment. So he accepts it and moves on.
