The entirety of the movie and the novel it was based on is exploring the importance of death, loss of innocence and self-discovery in a world where you're not fully understood along with sacrifice. The last Unicorn didn't truly understood humanity and the emotions that come with it, so during her journey when she was briefly a human, she's subjected to feeling both mortality and human emotions that unicorns don't and never experience. In a way, the movie is about how she discovers her full identity and learns how human emotions have their purposes and how important life can be. According to the original story, unicorns only understand the basic of emotions, but not the more complex ones. And on top of that, they're considered symbols of innocence and purity (whichever comes first) along with being (supposedly) wise, but they're, for lack of better term, detached from the outer world. Compared to how she was at the start of the film, the unicorn in the end is wiser and has a better understanding of humans and the emotions they can feel.
