ZP Developers Zone 16 June 2024
A weekly webinar from ZP answering questions for the week:
1) QUESTION ONE - …wire sensors
2) QUESTION TWO - making immunosensors
3) QUESTION THREE - Making a custom SPE
Question One
1. Can the wire biosensors operate at 3.3 V rather than 0.65 V as normally recommended per ZP specifications? Would this alter the calibration curve or the sensitivity of wire biosensor?
2. ATP wire biosensor is this wire sensor still available, also do you need glycerol for this sensor to work for the wire type?
3. Can we get a calcium wire biosensor? What would the cost of this one be for a pack of 4?
4. How many times can you use one of the wire biosensors in solution. Is it similar to the screen-printed electrodes by cleaning off with DI water?
5. Is calcium an amperometric or potentiometric sensor
6. What is the role of increasing voltage for the wire biosensors or screen-printed electrode? Will it help the sensitivity or the detection of the solution?
7. What is the range of detection for the Nitric oxide, ORP, hydrogen peroxide, ATP, Calcium for screen printed and wire sensors
8. What are the recommended current ranges to operate wire sensor: Hydrogen peroxide, Nitric oxide.
9. Do sensors need to be calibrated for each type of solution? For example, if you were to detect hydrogen peroxide added to pbs, DI water, or any other type of solution. Do each need their own calibration curve before they will function for that type of solution? Also, since the Nitric oxide sensor is difficult to calibrate yourself, does the calibration curve online serve as a good basis for measurements?
10. What type of liquid solution is best for calibrating sensors?
11. The calibration curves online, for each product, are these calibrations based on wire sensor or screen-printed wire sensors?
12. How does the Anapot device measure current in chronoamperometry mode based off the change in voltage?

Very nice presentations ZP on your channel and website. I'm not sure if my experiments are not going right because of enzymes or using electroanalytical methods wrongly. In the sensor I'm working on, I need to drive the reduction of Ferri to Ferro with a CA to measure the concentration of the substrate of some electroactive protein, in a process where FeCN should be the final electron donor. As I understand, the WE would be the source of electrons to drive this reduction. I'm able to get such a duck shape for my FeCN CV, am I correct to think that my CA should be set at a potential more negative than the potential of the reduction peak of FeCN? does it matter if I set it slightly positive (~50mV) of the reduction peak potential? Thanks in advance.

Hello Martin, If I understood the concept, to measure the EIS, the main input is voltage and current. Output will be complex impedance with respect to frequency. Could you pls agree with it? I am not technical background but looking forward your response. Is this measurement can be consider as safety?

    • @zimmerpeacock9703

"Just a quick TH-cam response" I found this just scrolling and you just blew my mind! Talking about Capsaicin, I'd just go HPLC-UV and problem solved. For context, I work in analytical chemistry. I do use voltametry with known and widespread techniques. But I have never heard of the possibilities you've just mentioned! Using the voltametric "spectrum" of a molecule of interest to get more specific with a "simple" voltametric measurement instead of the easy but sure way of chromatographic separation followed by any possible detection method is very interesting. Sure, you don't get the specificity of an HPLC, but the time and cost savings (an HPLC costs around 100 K $ and an analysis will take a few minutes uo to an hour depending on the specific matrix and goal) are pretty substantial! Is this actively used in a field I don't know, like medical analytics, or not and you're researching it to find an application? I'd guess the second, because I would guess that there are a lot of factors interfering with the voltammogram and potential ways to use it as a detection method. The usual method (we now use in chromatography, since we're separating the compounds before detection by chromatography) would be to just use the voltage of the first peak (normal method in HPLC-ECD). You can't really do that in potentiometry or voltametry or amperiometry, because other molecules will also get oxidized and give a response, though with different response factors or efficiency, which is why chromatography is still the way to go. But what if we would just add a dimension and use a correlation to the amperiogram as signal? This may even work pretty selectively without chromatography but simply directly measuring! The same would be possible with UV/VIS spectra, but we didn't develop that, because the UV/VIS spectra of similar molecules is also very similar and usually not distinguishable. So no option. But this surely is similar with voltammograms of specific molecules. So maybe we could combine absorption specra with the voltammogram of a specific molecule and find an algorithm able to distinguish them. Maybe another dimension/measurement will be needed, but this could be a breakthrough in analytical chemistry, making chromatography obsolete in some use cases! Though beside the personal interest, I hope this breakthrough, if it happens, won't happen to fast, because it would make a very expensive room full of chromatography equipment at the company I work in obsolete 😂 But scientific breakthroughs don't happen that fast. And after that there's also governments and their incredible inefficiency 😂😢 Which you can never trust, except for its talent to be centuries behind the state of the art 🙄 And other negative points we shall not mention in public 😂

Hello! I have a question of you don't mind. If a medical device manufacturer is ISO 13485 certified as part of the MDR requirements, does that mean that all the company's products should adhere to its QMS? Sir where can we get a screen printed electrode?

Thank you for this great explanation, and I appreciate very much the inclusion of the explanation of why use a three-electrode system! One comment/question: In the diagram that you provide you connect the reference electrode directly to the inverting terminal of the OpAmp. But, then, I think that the feedback voltage is the reference electrode's constant voltage, not the changing working electrode's voltage. I think I understand now from piecing a few different videos together that there should be a low-gain differential OpAmp that outputs the difference between RE and WE, and then that differential output is fed into the feedback.

That is the question at the heart of electroanalytical science, post it here and the tech team will respond - www.zimmerpeacock.com/contact/ -
