Since CpG and MDP have been shown to synergistically interact,- (TLR9 and NOD2 in Crohn's are lost),- maybe MDP,- just as it has been suggested with CpG,- could also be dependendent on p38 mapk-activated phosphorylation,- possibly located in the T-cell (CD40L) mediated help for APC maturation and IL-12p70 production? (Unmethylated) CpG activates innate immunity in a p38 mapk-dependent manner,- maybe MDP, which is related to CpG DNA, also activates p38 mapk phosphorylation and leads to transition to pMDP? In PBMCs, LPS or TLR4 as first signal for innate activation, then early IFN-gamma as next for irf1/IL-12p40, and lastly CD40L signal which is p38-dependent as third signal back in APC for full IL-12p35/70 production... leading to both early and late p38 mapk phosphorylation which might also enable nuclear import/export of specific molecules....

It's noticeable that it is possible to compare health state of fish in certain type with other kinds of gut action in case of infected with disease by gauge immunological parameters.
Thank you, for shedding light on the complex regulations taking place in the thymus! Concerning the last question, could CpG DNA be driving the innate IFNs from mTECs independently of both IFNARs? Have you looked at the mice model with IL12RB2 KO where KO mice develop spontanious autoimmunity, cancer and SLE/Sjögren's like changes/systemic vasculitis, in which a severe defect in Treg function has been shown pointing to a thymus defeciency? Interestingly, both AIRE and IL12RB2 is expressed in mTECs. In contrast to the only described family with a mutation leading to MSMD (IL-12/STAT4/IFN-g axis) due to a premature stop codon on exon 4 without showing any signs of complex immuno-defeciency and -dysregylation, in the IL12RB2 KO mice a broader deletion is induced including the promoter/enhancer/intronic regions, which could cause aberrant epigenetic modifications due to defecient upregulation of IFN-alpha/IL-2 induced upregulation of IL12RB2 and the functional synergy with IL-2/CD25/STAT5 activation and the modifications in the epigenetic landscape taking place during TCR activation and APC-T cell crosstalk (CD40Ligand-dependent). JMJD3 binds to 3 locations on the IL12RB2 intronic region and is inhibited by JAK2 inhibitor suggesting IL12RB2 dependency, which in the context of IL12RB2 defeciency could lead to both demethylation-dependent and -independent alterations,including in the shown co-operation between JMJD3 and T-bet-mediated SWI/SNF complex. Interestingly, IL12RB2 has also been linked to recruitment of p38 mapk and a defeciency of either one could lead to aberrant methylation status at CpG DNA (TLR9 pathway but from within, therefore independent of IFNARs) or of p38-mediated STAT1 signaling mechanisms. CpG methylation status could be driving IFN signature in mTECs expressing AIRE and IL12RB2 including in co-operation with IL-13-induced regulation of cellular 15-lipoxygenase activity on pretranslational, translational, and posttranslational levels which has been shown to be p38 mapk serine-dependent...
