could you please tell me what is th scatter sample that I should put it in the stage of IRF measurening
To measure the Instrument Response Function (IRF) of the FluoTime 300 system, you typically need a scatter sample that does not emit fluorescence. A common choice is a dilute colloidal suspension of nanoparticles like Ludox (colloidal silica), which efficiently scatters light but does not fluoresce. For example, take a drop of commercially available LUDOX TMA colloidal silica, 34 wt. % suspension in H2O, CAS Number: 7631-86-9 and dilute it 100 times with pure water. Can you please tell me whether a single photon is incident on the superconducting nanowires or a stream of photons fall on the nanowires for parallel nanowires setup?????
Thank your for your question. Both scenarios can happen, it depends on the signal intensity falling to the detector. These nanowire detectors were originally developed for real single-photon counting. The second half of Xiaolong Hu's presentation deals with detectors for multi-photon counting. Hi. To measure hydrophobicity with FLIM you need a dye with different quantum yield in water compared to hydrophobic media. You can google "NBD dye". The beam after pinhole is collimated an then focussed on the detector. The reason is that the active area of the detector is typically small

Should I use the same filter sets as my sample or should I excite the IRF “sample“ at the emission wavelength of my sample?
For measuring IRF in ideal case you should use the same emmision settings (for example filterset or wavelenght, intensity) and the same excitation settings (for example wavelenght, laser power, laser repetition rate) as for your sample. For real measurements, depending on the set-up type (spectrometer/microscope) or components (TiSa laser, diode laser, SPADs, PMT), variations of this general rule could be possible. Hi, thank you for the video! I wanted to ask if we should measure the IRF after (or before) every experiment or doing once for each filter should suffice? I am using a tunable 2p laser to excite and collect from either Turquoise (810 nm) or Ruby (1000 nm) and the filters I am using for detection are either 470/40 or 625/90. Is erythrosineB suitable for IRF of the Turquoise experiments? thank you!

Thank you for your comment. We will consider it and maybe we will make a video of it. PicoQuant offers the high performance luminescence spectrometer FT300. We can show you some performance tests of the FT300, but we don't know the properties of other spectrometer.

Hello picoQuant: I got an exponential decay curve of 14. k3V from 57Co radioactive source. My curve looks similar to the blue curve as in 2:00. I am curious to know how to fit the data using a exponential decay, with the vertical line found around 5 (xaxis)
