would be able to help me clear my doubts that what factors decide the type of calibration either linear or logit calibration? and also what determines number of standards required for an assay calibration?
The calibration line should be determined by the model you expect the data to fit. For absorption spectroscopy we use Beer's law as a model (A = ebc) which is a linear model so therefore the fitline of the calibration curve should be linear (As concentration increases, Absorption increases proportionally.) The number of points will determine the precision of your calibration. For a linear fit you would need no fewer than 3 points, but the more data points you include, the more precise the trendline is (the smaller the standard deviation).

Hello Teresa I have been using this method to measure the contact angle. I am wandering about the accuracy of this method. Can you please tell me about its accuracy and tolerance ?
The accuracy and precision depends on the user and the quality of the image of the drop. It is recommended that you use images of standard drop angles to calibrate your assessment, but also modifying how you take photos of the drops to get good contrast can help quite a bit.

Thank you Teresa. The video is pretty illustrative. I have a question. I'm working on a data set with 5 different independent experiments in which I got 3 measurements of each concentration of a drug and I registered several concentrations. It fits with a non linear regression. Shall I apply de G test to all the measurements of the same concentration or just to the triplicates in each independent experiment? Or maybe because of its distribution should I apply another method? Than you in advance and congrats for the video

your video is soo good, thank you for upload material like this. But I have a question, I work with Spartan 14 and the calculation table isnt the same and don't have the same options, I don't know what to do
I am using the student version here, so I'm sure the full version has a lot more options (and every student version is a bit different). Hartree-Fock and Density Functional are not necessary to get this animation, the semi-empirical option (PM3 in this case) is sufficient. I have run it at various levels of theory and the accuracy of the activation energy does not really improve, it's off by at least an order of magnitude regardless.
