    "Your study was presented very clearly. Although the data may not be representative of a larger population, it does not necessarily mean that the data will not be "reliable" (~8 min 30 sec). As long as your conclusion is specific to this study and discuss about the changes in bacteria after treatment in the test subjects

Do you think there might be any other forms of contamination from the environment during the packing and distribution process that could affect the reliability of your data and interpretation?

    This study is very relevant because many people may frequently leave out their open packs of kimchi. At around 6 minutes and 50 seconds, it was mentioned that the different fermentation times of various batches/packs of kimchi is a limitation. So the question is, why did you choose to test different batches of kimchi? Isn't that for better sampling?

    This is an interesting study. How would you connect your data to the effect of yogurt on eczema development? Do any of the test subjects have eczema? If you do obtain the expected results, such as increased S. thermophilus & L. bulgaricus and reduced S. aureus, is that enough to support the claim that yogurt can have a positive impact on eczema?

    Great presentation! Did you store the samples at -20 degrees Celsius or -80 degrees Celsius before DNA extraction? Do you think that is sufficient to preserve the sample integrity during storage, or would adding stabilizers help? Have you controlled for the skin surface area that is treated or swapped between individuals?

    By removing the soil sample from its natural condition, the microbiome may become less resilient to changes in pH. Given more resources and time, can you think of an experimental set-up that would allow you to assess the effect of pH on the soil microbiome outside?

    When it comes to gut health, "beneficial" bacteria are commonly isolated from fermented/dairy products. Does the literature suggest that these bacteria can similarly be isolated from agricultural products (specifically mushrooms)? If not, what beneficial bacteria do you expect to find on the organic mushrooms?

      Great question! While beneficial bacteria are commonly associated with fermented or dairy products, recent studies have found that mushrooms, especially organic varieties, can also host beneficial microbes. The literature points to bacteria like Bacillus, Lactobacillus, and even some beneficial strains of Pseudomonas, which are known to promote plant health and may have potential gut health benefits. Interestingly, the Happy Shroom Lab has observed a surprising decrease in both species richness and evenness in organic mushrooms compared to inorganic ones, which challenges initial expectations. Stay tuned as we continue to explore the curious case of the Happy Shroom Lab's mushroom microbiome misadventures!

    Nice presentation! Am glad you pointed out how this is an unnatural/abrupt pH change and other limitations might impact the conclusions that can be drawn. A little amused that you use a sterile shovel to dig soil haha!

    Nice videography - I notice you filmed from morning to night! A for effort!! I wonder if instead of drinking 1 full can, if the impact would have been more pronounced if you held a mouthful or herbal tea in your mouths for 2 minutes straight instead - but it was good that you thought about not using a straw! I also wonder if the temperature of tea (presumably most herbal teas would be taken warm/hot) would make a difference

As for your question, yes, inorganic mushrooms typically require more synthetic fertilizers and pesticides to support their growth. These help to boost nutrient availability and manage pests, ensuring higher yields. In contrast, organic mushrooms rely on natural fertilizers and biological pest control. This difference is one of the main factors the Happy Shroom Lab thinks influences the distinct microbial profiles between organic and inorganic mushrooms.

    Well done, group1! Interesting and engaging video that gets us all curious about the results! Just wondering, what is the rationale of swabbing after shower and not before? They may have been killed by preservatives, but wouldn't the DNA from these "probiotics" still be present at higher levels anyways? Finally, is there a better way to decide on study sample sizes instead of just following another study that has been published? Just because it is published doesn't make it right/good =)

    Why was this bifidobacterium strain used and not other strains? (Bifidobacterium bifidum BGN4 and Bifidobacterium longum BORI)

    It’s mentioned that there is a significant decrease in pro inflammatory genera within the probiotic group. However, the authors do not appear to have conducted any statistical analyses comparing abundance in the probiotic and placebo groups (which in some cases appear quite similar). Further, the relative abundance of these genera periodically decreases and increases regardless of treatment. Do you think the conclusion that probiotic supplementation beneficially influenced gut microbial communities is still valid?

    In the bioinformatics portion, you mentioned that Amplicon Sequence Variants (ASVs) were used for taxonomic identification. Why is that so? There are many other studies that used Operational Taxonomic Units (OTUs) (such as group1's study). What is the difference?

    What are your thoughts on this experiment translating to different ethnic patients clinically diagnosed with depression? How do you think their gut biota would be like? How would the results change? Good job on the presentation! Just some formatting suggestions: - take note of the speaker screen at the top right, sometimes it blocks out your words on the slides -some slides are too wordy. Might want to consider using graphics like flow charts and etc to reduce words - for results, if the figures from the paper does not add value then i suggest it can be replaced with (self-made) graphics instead??? Cause they are too small and I can’t and don’t really know where to see 😢 you can use it but perhaps would be good to highlight certain important results for viewers (box it up, highlight, pop-out box.. etc) Thank you for an informative presentation. I was wondering if the group could provide more clarification on what was meant by the study limitation, "deviations from the disease phenotype (DSS) does not necessarily imply a healthier state or rescue". What would deviations from the disease phenotype mean in this case then? Could they also have different but still negative impacts on health?

    Thanks for this interesting video! I found this study quite interesting as I think potential biomarkers are really needed in the context of ALS provided the large time gap between disease onset and eventual diagnosis! As ALS is often considered a rare disorder, I was wondering what you guys think can be done to incentivise researchers/companies to conduct further studies (as you described at the end) into microbiome dysbiosis as a diagnostic tool for rare diseases like ALS?
