Precisionary Webinar: Christopher Vaaga on "Neural circuits underlying innate fear habituation"

Dr. Christopher Vaaga is an Assistant Professor in the Department of Biomedical Sciences at Colorado State University. Dr. Vaaga started his research lab in 2022, after completing his graduate training at the Vollum Institute at the Oregon Health and Science University and postdoctoral training at Northwestern University. His research focuses on elucidating the neural circuit mechanisms underlying ethologically relevant behaviors, including understanding how these circuits are disrupted in various disorders. Specifically, his lab is examining the circuitry underlying innate fear responses, using a highly integrative approach spanning cellular intrinsic biophysics, in vitro slice electrophysiology, in vivo circuit manipulation and neural recording, viral circuit mapping, as well as behavioral analysis of fear responses. In this webinar, Dr. Vaaga will outline his lab’s recent efforts to understand how the cerebellum contributes to innate fear processing through direct synaptic interactions in the periaqueductal gray, a midbrain region essential for driving innate fear responses. He will then present new data from the lab characterizing important differences in the synaptic dynamics of two circuits within the periaqueductal gray.
In this webinar, Dr. Vaaga will:
1. Articulate the evidence supporting the role of the cerebellum in modulating innate fear circuits through synaptic interactions in the vlPAG.
2. Describe how innate fear responses change with repeated exposure and identify the environmental and physiological factors that impact this process.
3. Outline the differences in synaptic dynamics in the ventrolateral and dorsomedial periaqueductal gray.

    Hi maam, Jatin here. I want some guidence from you regarding liver tissue cutting in Leica cryotome. I am doing sectioning of rat liver tissue on cryotome but some cracking are their on tissue.     what are the pros/cons comparing a compresstome to a vibratome?

      Hi David, That's a great question! A Compresstome and a Vibratome share a lot of similarities. Primarily in that these machines, all operate as vibrating microtomes, allowing for various changes in the speed and frequency of oscillation of the blade. The main area that we begin to separate has to do with the embedding process and compression effect that you've seen on display in this video as well as many of our other ones. The specimen tubes, with their special compression lip, provides a measure of added stability to the tissue sample during the cutting process. This compression effect allows the Compresstome to section at speeds up to 5x faster than other vibrating microtomes and helps maintain structure morphology during the slicing process. This means that the Compresstome has been shown to cause less damage to surface cells and structures, which in turn leads to a higher healthy/dead cell ratio, better patch clamp recordings, a significant reduction in chattermarks and slice surface clarity, and also allowing the machine to cut some fixed tissue samples into the 4-7 micron thickness range! The Compresstome has been used by scientists and institutions around the world for over 14 years now due to the very real benefits this compression method can provide. The Allen Institute for Brain Science has been one of our biggest supporters over the years and has been using the Compresstome to provide excellent acute brain slices for patch clamp recordings.       Hi! Thank you for your question. Our Compresstome VF-200 series and VF-300 series models have customizable specimen tube diameters. Since the cutting blades and blade holder themselves are of a fixed size, this means that as the size of the tube grows larger, the angle of approach for the blade itself must be increased in order for the blade to properly cut across the entire face of the tube. Sometimes when customers end up purchasing a larger sized specimen tube from us, they see that the blade might not cut across the entire sample. This video is meant as a visual aid for those customers. The majority of customers will never need to adjust their instrument in this fashion. I hope this helps answer your question!
