Do CAR-T Cells Cause or Become Cancer? Episode 025 of For Whom the Cell Tolls Podcast

The basic answer is no. When faced with R-CHOP resistance, DLBCL patients now have the option to utilize genetically modified Chimeric Antigen Receptor T-cells (CAR-T) designed to hunt tumors. This episode investigates the recent controversy into if these super-cells can transition into a villainous tumor themselves. We cover a paper by Garcia et. al. that showcases how much power we are adding to CAR-T, even activating oncogenes to do it. Next, we transition to a large analysis from Stanford where 724 patients are examined, with 1 developing a T-cell tumor. Sequencing this case reveals that no synthetic vector DNA integrations or activity is within the tumor. It ends up revealing a great surprise about shared evolutionary pathways between B & T-cell progenitor stem cells. Enjoy the episode!

    Thank you Keenan great video. I was trying to figure out why I didn't have the nice data when I put >2 rows/columns in contingency data. To better assess where the statistical significance arrises from - Can I separately analyse 2 x 2 using fishers etc; using different combinations of different groups i'm analysing

      Hello! The contingency format is super rough looking in terms of visuals sadly. I break my "no pie graphs rule" when the data is only 2x2, making two, but then I'll perform the Fisher's separately. If you deviate from 2x2 format of data, we typically don't use Fisher's although this is mainly because it's tough by hand. Since that's less of a problem with computers, you can, but I've seen Chi-Square applied more often when it goes beyond 2x2. You can run as many separate Fisher's as you need if you have the data, but there may be some kind of multiple-hypothesis correction to apply separately in that case. Hope this helps!

    Hello Keenan, is there a way that you can change the shape of the symbols to show the categories in different ways. Like in you example 1, 2, 3 etc would be a square, triangle and circle?

      Hello! Prism's XY format would definitely allow for that if you were okay with escaping the Multiple Variables format. The MV settings are a bit clunkier, but there still should be a scroll down menu to choose which data (and axis most likely) different shapes could apply to. Search about, and let me know if you find a path!
